description: JANUMET is a drug used as a diabetes medication. Janumet contains metformin and sitagliptin. Metformin is a type 2 anti-diabetic drug which belongs to the biguanide group. Metformin works by suppressing glucose production by the liver, increasing insulin sensitivity, increasing peripheral glucose absorption, and reducing insulin-induced suppression of the fatty acid oxidation process. Sitagliptin is an anti-hyperglycemic agent from the class of dipeptidyl peptidase-4 inhibitors. Sitagliptin inhibits the inactivation of GLP-1 and GIP by DPP-4, which allows GLP-1 and GIP to potentiate insulin secretion in beta cells and suppress glucagon release by alpha cells of the Islets of Langerhans in the pancreas. The intense increase in insulin ratio caused by increased levels of incretin hormones results in a decrease in fasting and postprandial hepatic glucose production. This causes reduced glycemia.
